RESEARCH EXPERIENCE AND PROFESSIONAL TRAINING:

2003-2005 David Geffen School of Medicine at UCLA, Department of Urology, Urologic Oncology Laboratory, Los Angeles, California.
 Position: urologic oncology research fellow
 Principal Investigator: Arie S. Belldegrun, MD.
Research involving the evaluation of genetically engineered dendritic cells expressing carbonic anhydrase IX (CA IX) or CA IX-granulocyte/macrophage-colony stimulating factor (GM-CSF) fusion protein to generate protective and active immunity for the prevention and treatment of tumors expressing CA IX and defining the immunological mechanisms governing CA IX-DC generated immunity.  Research also involving the development of second-generation tissue microarrays to provide prognostic information on patients with advanced kidney cancer and identifying those patients most likely to benefit from CA IX-directed therapy.  Research also involving the use of hypoxia-inducible factor-1a (HIF-1a) as a potential target for immunotherapy of renal cell carcinoma.

Research involved gene modification of antitumor T cells for immunotherapy of cancer patients.  Research efforts demonstrated receptor expression level is an important factor in retroviral-target interactions and gene transfer efficiency in human lymphocytes was improved 4- to 18-fold over previous gene transfer protocols using the gibbon ape leukemia retrovirus.  A novel quantitative polymerase chain reaction (PCR) assay was developed to determine transduction efficiency.  In addition, studies involved use of tumor antigens in the development of potential therapeutic vaccines.  Research efforts demonstrated dendritic cells pulsed with tumor antigens or recombinant poxviruses encoding tumor antigens were effective in prolonging survival and treating established pulmonary metastases in a murine tumor model.

 Principal Investigator: Robert M. Merion, MD.
Research efforts demonstrated that portal venous inoculation (PVI) of donor strain mononuclear cells into recipient animals significantly inhibited mixed lymphocyte culture reactivity (MLC) and liposome-mediated reticuloendothelial system (RES) blockage returns proliferation of lymphocytes back to control levels of MLC reactivity.  These results suggested that an intact RES is necessary at the time of PVI in order for inhibition of MLC to occur.
